چکیده

Allergic airway inflammation and asthma have emerged as major public health problem in the Midwest, United States of America. Targeting Toll-like receptor (TLR) signaling and Calcium-activated Chloride (ClCa)/Potassium (KCa) Channels in unraveling the cellular/molecular mechanisms underlying genetic susceptibility to inflammatory diseases in specific human patient population subset(s) is emerging as an attractive immunotherapeutic pharmacological strategy in management/prevention of asthma. Objectives: My pilot study aimed to investigate the role of TLRs and ClCa/KCa ion channels in human bronchial epithelial cells (NHBEC, BEAS-2B from ATCC), and eosinophils-derived from asthma patients of North American ethnicity. Methods: Whole cell patch clamp electrophysiological recordings were conducted for ClCa and KCa currents in cultured cells (passages P3-P7) grown on cover-slips. RNA and Protein were extracted from NHBECs, BEAS-2B and eosinophils using Trizol and RIPA methods. Pipette and bath solutions for electrophysiology were subjected to stringent pH and osmolality checks prior

Overview of Relationship of Allergic Asthma with Diet and Food Intake

Allergic asthma is the most common acute inflammatory airway disorder and their incidence population has exploded in the last years, being estimated that over 300 million people worldwide have the condition. Here, we present a brief overview of relationship of allergic asthma with diet and food intake. Several studies have shown that most asthma cases are due to an allergic immune response to e...

Inactivated Mycobacterium phlei inhalation ameliorates allergic asthma through modulating the balance of CD4+CD25+ regulatory T and Th17 cells in mice

Objective(s): Th2 response is related to the aetiology of asthma, but the underlying mechanism is unclear. To address this point, the effect of nebulized inhalation of inactivated Mycobacterium phlei on modulation of asthmatic  airway  inflammation was investigated. Materials and Methods: 24 male BALB/c mice were randomly divided into three groups: control group (Group A), asthma model group (G...

The Role of Toll-Like Receptors in Breast Cancer

Breast cancer is highly prevalent worldwide. Treatment of this disease is a major global public health problem; therefore, numerous studies have explored new molecular profile helping for breast cancer therapy. Different risk factors are attributed to breast cancer; however, chronic inflammation and immune regulation are highly essential. Various molecules impact this context, including toll-li...

Cleavage of fibrinogen by proteinases elicits allergic responses through Toll-like receptor 4.

Proteinases and the innate immune receptor Toll-like receptor 4 (TLR4) are essential for expression of allergic inflammation and diseases such as asthma. A mechanism that links these inflammatory mediators is essential for explaining the fundamental basis of allergic disease but has been elusive. Here, we demonstrate that TLR4 is activated by airway proteinase activity to initiate both allergic...

Background: Allergic Asthma is an inflammatory disease of the respiratory system that is well known by increased inflammatory cells in the airways and causes difficulty in respiration. The prevalence of allergic asthma is increasing worldwide, and it has become a significant cause of health challenge especially in developed countries. Inhaled β2-agonists and Inhaled or oral corticosteroids are ...
